Can I use DNAbsolute to extract DNA from solid samples?
Yes, DNAbsolute is effective in isolating DNA from dried samples. So far, DNAbsolute has demonstrated efficient extraction of DNA from desiccated corals and plants (either herbarium specimens or fresh material desiccated by a freeze-drying or silica gel method).
How does DNAbsolute work?
DNAbsolute is composed of an ionic liquid that has a selective and high affinity for DNA strands. When mixed with the sample, DNAbsolute reagent binds directly to DNA and precipitates it without the need for RNase treatment, protein precipitation or the use of hazardous reagents.
Can I use DNAbsolute to recover high MW DNA fragments?
Yes, DNAbsolute is effective in the recovery of HMW DNA fragments (larger than 48kb for dry corals, larger than 24kb for arthropods).
How pure will the DNA extract be?
The DNAbsolute method can produce high purity DNA due to its ability to efficiently and specifically solubilize DNA, minimizing contamination with proteins, RNA and other cellular debris.
For example, the purity of drosophila and coral DNA extracted with DNAbsolute was assessed by spectroscopic measurements. The average 260/280 ratio was found to be between 1.8-1.9 and the 260/230 ratio at 2.3.
The type of biological sample being processed can affect the purity of the extracted DNA. Optimization of sample lysis, or additional purification steps, could be implemented when working with samples containing high levels of secondary metabolites.
How long does it take to extract DNA with DNAbsolute?
Once the sample is lysed, the entire DNA extraction process takes about 30 minutes.
What performance can I expect?
The resulting yield can vary greatly depending on sample type and preparation. As a general rule, an average of 85% of DNA can be expected to be recovered from a solution ranging from 10 to 100 μg/mL DNA, and 60% from a solution ranging from 5 to 10 ng/mL DNA. If the starting material is of poor quality and/or fragmented, the yield of purified DNA will be lower.
