Enzyme Immunoassay for the quantitative determination of Ibrutinib in serum, plasma and cell culture supernatant Ibrutinib (Imbruvica) is a small molecule drug that binds permanently to a protein, Bruton's tyrosine kinase (BTK), and it is important in B cells. It is used to treat B cell cancers like mantle cell lymphoma, chronic lymphocytic leukemia, and Waldenström's macroglobulinemia. About the kit: - Uses anti-idiotypic antibodies sourced from our vendor partner in the US, which ensures higher specificity, and low cross reactivity. - Recovery rates are between 85 - 115% - Ready to use with a standard protocol with break-apart pre-coated wells - Validated as per US FDA guidelines for Bioassays - Optimized for matrix effects to ensure higher sensitivity. - Shelf life: 1 year The Ibrutinib ELISA is a competitive immunoassay for the determination of Ibrutinib. A constant concentration of Tracer; varying concentrations of unlabeled standard and BTK are added in sequence to the uncoated microtitre plate wells and incubated. After 1st incubation, Anti- BTK Antibody is dispensed to all wells of uncoated Microtitre plate containing the reaction mixture and incubated. After 2nd incubation, all reaction mixtures are transferred to the respective wells of coated microtitre plate and incubated. After washing, Streptavidin HRP conjugate is added and incubated. Captured Tracer is subsequently bound by streptavidin HRP which produces a soluble colored product after addition of TMB substrate. The enzyme reaction is stopped by dispensing of stop solution into the wells. The optical density (OD) of the solution at 450 nm is inversely proportional to the amount of bound Ibrutinib molecule present in standards or samples.
