Immunoassay for Quantitative Determination of Human IgE in cell culture supernatant and other biological preparations Immunoglobulin E (IgE) is a type of antibody (or immunoglobulin (Ig) "isotype") that has only been found in mammals. IgE is synthesised by plasma cells. Monomers of IgE consist of two heavy chains and two light chains, with the chain containing 4 Ig-like constant domains. IgE's main function is immunity to parasites such as helminths like Schistosoma mansoni, Trichinella spiralis, and Fasciola hepatica. IgE is utilized during immune defense against certain protozoan parasites such as Plasmodium falciparum. About the kit: - Uses anti-idiotypic antibodies sourced from our vendor partner in the US, which ensures higher specificity, and low cross reactivity. - Recovery rates are between 85 - 115% - Ready to use with a standard protocol with break-apart pre-coated wells - Validated as per US FDA guidelines for Bioassays - Optimized for matrix effects to ensure higher sensitivity. - Shelf life: 1 year This assay is based on the Sandwich ELISA procedure. Samples containing IgE react with already coated affinity purified capture anti-Human IgE antibody and bind to them. Plates are washed with wash buffer to remove unbound reactants. Biotinylated Anti-Human IgE is added leading to formation of a sandwich complex of solid phase antibody-Human IgE-enzyme labeled antibody. The wells are washed to remove any unbound reactants as per the wash procedure. Streptavidin: HRP conjugate is added which binds to Biotinylated Anti-Human IgE complex. The wells are washed to remove any unbound reactants as per the wash procedure. The substrate 3, 3’,5, 5’ Tetra Methyl Benzidine is then reacted. The amount of hydrolyzed substrate is read on a microtiter plate reader and it is directly proportional to the concentration of Human IgE present in the samples.
